Myocardial Actions of Angiotensin

چکیده
• The cardiovascular effects of angiotensin have been studied extensively since its identification 25 years ago." The synthesis of various angiotensins and the demonstration of the pharmacological identity of the naturally occurring and the synthetic octapeptides have facilitated such studies in recent years. It has been found consistently that angiotensin is a powerful constrictor of arteriolar smooth muscle, increases total peripheral resistance, and raises the systemic arterial pressure. No such agreement exists concerning the actions of the drug on heart muscle. Of 32 published studies on changes in the normal human or animal circulation following administration of angiotensin, 19 have found that cardiac output is decreased and 13 that it remains unchanged. Increases of diastolic volume of the heart, of left ventricular end diastolic pressure, of diastolic pressures in the other cardiac chambers, of pulmonary wedge pressure, of central venous pressure, and of circulation time have also been observed following introduction of angiotensin into the intact circulation.^ These observations have led some workers to conclude that angiotensin depresses the strength of contraction of heart muscle or that it lacks inotropic action on the heart.'-On the other hand, studies in which myocardial contractility was evaluated more directly suggest that at least some concentrations of angiotensin exert positive inotropic effects on mammalian myocardium. St ~Some of these reports describe only a few observations or yield no quantitative data; the increases in contractility observed in the other studies

Effect of losartan on NOX2 transcription following acute myocardial ischemia-reperfusion
Introduction: Nicotinamide adenine dinucleotide phosphate (NADPH) oxidase-2 (Nox2) is one of the predominant sources of ROS production during myocardial ischemia-reperfusion and can be induced by angiotensin II. The evidence suggests that pharmacological blockers of renin-angiotensin system can exert direct tissue effects independent of their ability to regulate blood pressure. The mechanism...
متن کاملEffects of pretreatment with non hypotensive dose of ramiprilat and losartan on myocardial ischemia-reperfusion induced arrhythmias and infarct size in rats
Introduction: Inhibition of renin angiotensin system represents an important approach in the management of cardiovascular diseases. The aim of this study was to explore the effects of pretreatment with non-hypotensive dose of angiotensin converting enzyme (ACE) inhibitor, ramiprilat and angiotensin type 1 (AT1) receptor blocker, losartan on myocardial infarct size and arrhythmias in a rat mo...

In this study, the changes of renin system activity and the effects of aspirin therapy on the plasma renin activity ( PRA) in myocardial infarction were investigated. Fifty nine patients with myocardial infarction were selected. By suppressed in the former group which was statistically significant ( p<0.01).

متن کاملContribution of bradykinin to the cardioprotective action of angiotensin converting enzyme inhibition in hypertension and after myocardial infarction.
Angiotensin converting enzyme (ACE) is identical with kininase II. Besides reducing the production of angiotensin II, inhibition of ACE potentiates the biological actions of endogenous kinins. In hypertension-induced left ventricular hypertrophy, potentiation of endogenous kinins contributes to the improvement of cardiac function and energy metabolism and to capillary proliferation effected by ...
متن کاملResveratrol Suppresses Cardiac Renin Angiotensin System in the Late Phase of Left Ventricular Hypertrophy
Background and objectives: Resveratrol(3,5,4′-trihydroxy-trans-stilbene) is a natural polyphenole phytoalexin which exerts potential cardioprotective effects, but the cellular and molecular mechanisms responsible for these effects are still unknown. Cardiac renin angiotensin system (RAS) over-activation plays an important role in pathogenesis of left ventricula...
